tsRNA-3040b antagomir is a chemically-modified, single-stranded antisense oligonucleotide designed to inhibit the activity of tsRNA-3040b, a transfer RNA-derived small RNA (tsRNA). Synthesized with phosphorothioate linkages and 2'-methoxy modifications to enhance stability and cellular uptake, this compound is used as a research tool to investigate the role of tsRNAs in disease. Its primary mechanism involves binding to tsRNA-3040b, which otherwise accumulates R-loops to regulate the transcription of Tripartite motif-containing protein 35 (Trim35). By inhibiting tsRNA-3040b, the antagomir modulates the Wnt3A/β-catenin signaling pathway and glucose metabolism (glycolysis), thereby reducing pulmonary artery endothelial cell (PAEC) proliferation. In preclinical models of hypoxia-induced pulmonary hypertension, the antagomir has demonstrated efficacy in improving cardiac function and reducing vascular remodeling.
