Overview

This is a combination therapy comprised of TST001 (osemitamab), gemcitabine, and nab-paclitaxel. **TST001** is a humanized IgG1 monoclonal antibody targeting claudin 18.2 (CLDN18.2), a tight junction protein aberrantly expressed in various gastrointestinal malignancies, notably gastric/gastroesophageal junction (G/GEJ) and pancreatic cancers. It exerts anti-tumor effects primarily through antibody-dependent cell-mediated cytotoxicity (ADCC), with enhanced NK cell activation due to reduced fucose content[5][1]. **Gemcitabine** is a nucleoside analog antimetabolite interfering with DNA synthesis and repair, and **nab-paclitaxel** (albumin-bound paclitaxel) is a microtubule inhibitor formulated to improve paclitaxel delivery and tolerability[2][6]. This triple combination is under investigation for HER2-negative, unresectable, locally advanced, or metastatic gastric, gastroesophageal junction, and pancreatic cancers.
