Overview
A four-drug combination regimen used as first-line therapy for unresectable locally advanced or metastatic gastric/gastroesophageal junction (G/GEJ) cancer. TST001 (osemitamab) is a humanized monoclonal antibody targeting Claudin 18.2 (CLDN18.2), with enhanced antibody-dependent cellular cytotoxicity (ADCC). Oxaliplatin is a platinum-based chemotherapeutic. Capecitabine is an oral fluoropyrimidine antimetabolite. Nivolumab is a programmed death 1 (PD-1) immune checkpoint inhibitor. The combination leverages targeted therapy (TST001), immunotherapy (nivolumab), and chemotherapeutic cytotoxicity (CAPOX = capecitabine + oxaliplatin) to maximize anti-tumor efficacy regardless of CLDN18.2 or PD-L1 expression. This regimen is under investigation in phase 1/2 clinical trials for efficacy and safety in advanced G/GEJ cancer[5].
