TST003 is a high-affinity, humanized monoclonal antibody that targets Gremlin1 (GREM1), a member of the TGF-beta superfamily. Gremlin1 is a secreted protein highly upregulated in multiple solid tumors and plays an important role in promoting epithelial-mesenchymal transition and tumor progression. By specifically binding to and neutralizing gremlin-1 expressed on tumor cells and cancer-associated fibroblasts, TST003 inhibits GREM1-mediated signaling pathways, including FGFR1 phosphorylation, which are implicated in cancer cell proliferation, invasion, and resistance to checkpoint inhibitors. Preclinical studies have shown that TST003 has potent single-agent anti-tumor activity in patient-derived xenograft models of difficult-to-treat solid tumors such as castration-resistant prostate cancer and microsatellite stable colorectal cancer. It also enhances the efficacy of checkpoint inhibitors in syngeneic tumor models. Developed by Suzhou Transcenta Therapeutics (Transcenta), TST003 has received FDA clearance for clinical testing and is currently being evaluated in Phase 2 trials for adenocarcinoma/colorectal cancer and Phase 1 trials for other solid tumors[4][3][6][7][8].
