**TST005** is an investigational, Fc-silenced bifunctional immuno-oncology fusion protein developed by Transcenta for advanced solid tumors. It combines a high-affinity humanized anti-programmed death-ligand 1 IgG1 antibody with the truncated extracellular domain of transforming growth factor beta receptor type II, which functions as a transforming growth factor beta trap. TST005 is designed to restore antitumor T-cell activity by blocking programmed death-ligand 1 interaction with programmed cell death protein 1 while neutralizing transforming growth factor beta-mediated immunosuppression and immune exclusion in the tumor microenvironment. It is being evaluated as an intravenous Phase 1 therapy in locally advanced or metastatic solid tumors, including an expansion cohort for human papillomavirus-related malignancies.
