TST786 is a trispecific antibody (TsAb) developed by Transcenta Holding Limited for the treatment of solid tumors, specifically colorectal and prostate cancers. It is designed to simultaneously target three distinct pathways: the Programmed Cell Death Protein 1 (PD-1) immune checkpoint, the Vascular Endothelial Growth Factor (VEGF) angiogenic pathway, and Gremlin-1 (GREM1), a bone morphogenetic protein (BMP) antagonist. By inhibiting PD-1, TST786 aims to reactivate anti-tumor T-cell responses; by targeting VEGF, it disrupts tumor angiogenesis; and by inhibiting Gremlin-1, it modulates the tumor microenvironment and potentially reverses epithelial-mesenchymal transition (EMT). The drug is currently in the IND-enabling stage of development.
