TSY-120 is a preclinical-stage antibody-drug conjugate (ADC) biosimilar developed by Formosa Pharmaceuticals. It is designed as a biosimilar to Enhertu (trastuzumab deruxtecan), targeting the human epidermal growth factor receptor 2 (HER2). The drug is intended for the treatment of HER2-positive and HER2-low breast cancers. TSY-120 consists of a humanized anti-HER2 monoclonal antibody covalently linked to a cytotoxic topoisomerase I inhibitor payload via a cleavable linker. By binding to HER2 on the surface of tumor cells, the ADC is internalized, and the payload is released to induce DNA damage and cell death, including a bystander effect on neighboring tumor cells.
