Overview

TT-00920 is an orally bioavailable, potent, and highly selective small molecule inhibitor of phosphodiesterase type 9 (PDE9), developed by TransThera Biosciences for the treatment of chronic heart failure. The drug is designed to be low brain-penetrant and acts by inhibiting PDE9A, which regulates natriuretic-peptide-stimulated cyclic guanosine monophosphate (cGMP) in cardiac myocytes. PDE9 is strongly upregulated in human heart failure, particularly in heart failure with preserved ejection fraction (HFpEF), making it a promising therapeutic target. By inhibiting PDE9, TT-00920 aims to restore the cardioprotective mechanisms that are dysfunctional in heart failure, improving cardiac function and reducing ventricular remodeling and fibrosis as demonstrated in preclinical models. The drug has completed Phase 1 clinical trials evaluating safety and pharmacokinetics in healthy volunteers[1][2][3][5][6].
