Overview

TT-10 is an investigational small molecule drug developed as an antagonist of the adenosine A2A receptor (ADORA2A). It is being studied primarily for its potential in treating various advanced solid tumors, including non-small cell lung cancer, renal cell carcinoma, castration-resistant prostate cancer, head and neck squamous cell carcinoma, colorectal cancer, endometrial carcinoma, and ovarian cancer. The drug acts by inhibiting the immunosuppressive effects mediated by adenosine signaling in the tumor microenvironment via blockade of the A2A receptor. This mechanism aims to enhance anti-tumor immune responses. TT-10 is administered orally and has reached Phase 2 clinical trials for several indications[3][4][7][8]. In preclinical research unrelated to oncology (notably in cardiac repair), TT-10 has also been shown to activate components of the Hippo-Yap signaling pathway—promoting cardiomyocyte proliferation and survival after myocardial infarction—but this application is distinct from its current clinical development focus[1][5].
