TT-10 + TT-4 is an investigational oral combination therapy consisting of two small molecule antagonists targeting the adenosine receptor pathway for cancer immunotherapy. Specifically, TT-10 (also known as PORT-6) is a potent and selective antagonist of the adenosine A2A receptor (A2AR), while TT-4 (PORT-7) is a potent and selective antagonist of the adenosine A2B receptor (A2BR). The dual blockade approach aims to inhibit both A2AR and A2BR signaling in the tumor microenvironment, which are implicated in immune suppression and tumor progression. This combination is being evaluated for safety, tolerability, pharmacokinetics/pharmacodynamics, anti-tumor activity, and preliminary efficacy in patients with advanced solid tumors who have failed or are not eligible for standard-of-care treatments[1][2][3][4].
