TT-3 is a small molecule antagonist of the adenosine A2B receptor (A2BR) currently under development by Cyncado Therapeutics. The drug is in the IND-enabling phase, specifically targeting gut cancers. In the tumor microenvironment, high levels of adenosine contribute to immunosuppression and tumor progression; A2BR activation is particularly associated with promoting angiogenesis, metastasis, and the suppression of anti-tumor immune responses in gastrointestinal malignancies. By selectively inhibiting A2BR, TT-3 aims to disrupt these pro-tumorigenic pathways and enhance the immune system's ability to clear cancer cells.
