Overview

TT-816 is a first-in-class, oral small molecule antagonist of the cannabinoid CB2 receptor developed by Teon Therapeutics for the treatment of a broad range of solid tumors. Acting as an immune checkpoint inhibitor and novel immune response modifier, TT-816 is highly selective for the cannabinoid CB2 receptor over CB1. Its mechanism involves inhibiting cannabinoid CB2 receptors on immune cells, which enhances T cell and natural killer (NK) cell activity, promotes T cell infiltration into solid tumors, prevents T cell exhaustion, and synergizes with anti-PD-1 therapies. Preclinical studies have shown that TT-816 increases tumor-infiltrating lymphocytes and has additive or synergistic effects with PD-1 inhibitors in both "hot" and "cold" tumor models. The drug is being evaluated in Phase 1/2 clinical trials as monotherapy and in combination with pembrolizumab (a PD-1 inhibitor) for advanced solid tumors including non-small cell lung cancer (NSCLC), ovarian cancer, renal cell carcinoma (RCC), and small cell lung cancer.
