**TT16** is an investigational multimodal immunotherapy for advanced **HER2-positive solid tumors**. It combines local intratumoral delivery of **CAdVEC**, a binary oncolytic adenovirus engineered to express interleukin-12p70 and a programmed death-ligand 1 blocking antibody, with subsequent intravenous infusion of autologous HER2-specific CAR virus-specific T cells. The adenoviral component is intended to lyse tumor cells and remodel the immunosuppressive tumor microenvironment, while the HER2-directed CAR-T-cell component recognizes and eliminates HER2-expressing cancer cells. TT16 was developed by Tessa Therapeutics in collaboration with Baylor College of Medicine and is being evaluated in the Phase 1 VISTA study in advanced HER2-positive solid tumors.
