TT3-LITE (Total Therapy 3-Lite) is a modified, less intensive multi-drug chemotherapy and stem cell transplant regimen developed by the University of Arkansas for Medical Sciences (UAMS) for the treatment of multiple myeloma. It was designed to improve the therapeutic index of the standard Total Therapy 3 (S-TT3) protocol by reducing treatment-related toxicities by approximately 50%, particularly for patients with low-risk disease or those who may not tolerate the full intensity of the standard protocol. The regimen typically incorporates induction therapy with agents such as bortezomib, thalidomide, and dexamethasone (VTD), often combined with cisplatin, doxorubicin, cyclophosphamide, and etoposide (PACE), followed by autologous stem cell transplantation and maintenance therapy.
