TTA-121

TTA-121 is a novel intranasal formulation of oxytocin developed by Teikoku Pharma, specifically engineered for enhanced bioavailability compared to conventional oxytocin sprays like Syntocinon. By optimizing its physicochemical properties, TTA-121 achieves approximately 3.6 times higher delivery to the brain, as measured by area under the concentration-time curve (AUC) in animal models. It is primarily being investigated for the treatment of core social symptoms in individuals with Autism Spectrum Disorder (ASD). Clinical trials have demonstrated an inverted U-shaped dose-response relationship, with moderate doses showing potential efficacy in improving social reciprocity scores. The drug functions as an agonist at the oxytocin receptor, a key mediator of social cognition and behavior.
