Overview

TTC-909 is an investigational lipid microsphere formulation of **isocarbacyclin methyl ester**, a stable analog of prostaglandin I2 (prostacyclin). Developed by **Taisho Pharmaceutical** in collaboration with **Teijin**, it was primarily evaluated for the treatment of **primary pulmonary hypertension (PPH)**, particularly in pediatric populations. The formulation utilizes a lipid microsphere (LM) delivery system, typically consisting of soybean oil and lecithin, with a particle size of approximately 0.2 μm. This delivery modality is designed to target the pulmonary vasculature, protecting the active moiety from rapid metabolic inactivation in the lungs and reducing systemic side effects such as hypotension. Isocarbacyclin acts as a potent agonist at the **prostacyclin (IP) receptor**, stimulating adenylate cyclase to increase intracellular cAMP, which leads to significant vasodilation of pulmonary arteries and inhibition of platelet aggregation.
