Overview

tTF-NGR is a recombinant fusion protein composed of the extracellular domain of truncated tissue factor (tTF) conjugated to a C-terminal NGR peptide (GNGRAHA). It is designed as a vascular-targeting agent that binds to aminopeptidase N (CD13), which is preferentially expressed on tumor vasculature and some tumor cells. Upon binding, tTF-NGR acts as an agonist of Aminopeptidase N, inducing localized thrombosis within the tumor blood vessels, leading to infarction and necrosis of the tumor tissue. This mechanism aims to selectively reduce blood flow in tumors without affecting normal tissues. The drug has been studied in combination with cytotoxic agents such as trabectedin for metastatic soft tissue sarcoma, with the goal of enhancing anti-tumor efficacy by trapping chemotherapeutic drugs inside tumors through vascular occlusion. Clinical studies have shown selective reduction in tumor blood flow and evidence of intratumoral hemorrhage and necrosis following administration[1][2][3][5][6][8].
