Overview

TTF1-NP is a nanoparticle formulation of 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone (TTF1), a bioactive flavone isolated from *Sorbaria sorbifolia*. Developed to overcome the poor solubility of the parent compound, TTF1-NP exhibits potent anti-cancer activity, particularly in hepatoma (liver cancer) models. Its mechanism of action involves the downregulation of Basic Transcription Factor 3 (BTF3), which disrupts the BTF3-HIF-1α complex. This disruption promotes the ubiquitination and subsequent proteasomal degradation of Hypoxia-Inducible Factor 1-alpha (HIF-1α), leading to the inhibition of glycolysis (the Warburg effect) and the induction of G0/G1 cell cycle arrest. Preclinical studies in mouse xenografts and rat primary liver cancer models have demonstrated that TTF1-NP significantly attenuates tumor growth, reduces lactate production, and decreases ATP levels, suggesting its potential as a novel dual-targeted therapeutic candidate for liver cancer.
