Overview

TTFC-E7SH is an investigational therapeutic DNA vaccine developed by the Netherlands Cancer Institute (NKI-AVL) for the treatment of HPV16-positive high-grade vulvar intraepithelial neoplasia (uVIN). The vaccine construct encodes a fusion protein consisting of the Tetanus Toxin Fragment C (TTFC), which acts as an adjuvant/carrier to enhance immune recognition, and a shuffled, non-oncogenic variant of the HPV16 E7 oncoprotein. It is designed to elicit a T-cell-mediated immune response against cells infected with HPV16. In clinical evaluation, the vaccine was administered intradermally using a DNA tattooing technique. While the Phase I study demonstrated a favorable safety profile, it showed limited immunogenicity and a lack of clinical efficacy, prompting the development of improved next-generation vaccine formats.
