TTFields + temozolomide is a combination therapy used primarily for the treatment of newly diagnosed glioblastoma. Tumor Treating Fields (TTFields) are low-intensity, alternating electric fields delivered non-invasively to the scalp via transducer arrays (commercially known as Optune). These fields disrupt mitosis and inhibit tumor cell proliferation by interfering with microtubule assembly and other cellular processes essential for cancer cell division. Temozolomide is an oral alkylating agent that induces DNA damage in tumor cells, leading to apoptosis. The combination has demonstrated synergistic effects in preclinical and clinical studies, resulting in improved progression-free survival (PFS) and overall survival (OS) compared to temozolomide alone. This regimen is approved as first-line maintenance therapy following chemoradiation in newly diagnosed glioblastoma patients[2][3][4][5].
