TTI-101 + palbociclib is a combination therapy being investigated for hormone receptor-positive (HR+), HER2-negative (HER2-) metastatic breast cancer, particularly for patients who have developed resistance to standard palbociclib therapy. TTI-101 is a first-in-class, selective small-molecule inhibitor of Signal Transducer and Activator of Transcription 3 (STAT3), while palbociclib is a CDK4/6 inhibitor. Research has shown that resistance to palbociclib is often driven by activation of the IL-6/STAT3 pathway, and inhibition of STAT3 with TTI-101 can overcome this resistance. The combination has demonstrated synergistic effects in arresting tumor growth in preclinical models.
