Overview

TTI-621 + nivolumab is an investigational combination therapy composed of TTI-621, a recombinant fusion protein (SIRPα-IgG1 Fc) that blocks the CD47 "don't eat me" signal, and nivolumab, a monoclonal antibody targeting the programmed death-1 (PD-1) receptor[1][3][7]. By simultaneously inhibiting both the innate immune checkpoint (CD47/SIRPα axis) and the adaptive checkpoint (PD-1), this combination aims to potentiate anti-tumor immune responses through enhanced macrophage-mediated phagocytosis and boosted T-cell activation[1][4]. TTI-621 is developed primarily for hematologic malignancies including Hodgkin lymphoma and cutaneous T-cell lymphoma, with preliminary studies showing safety and early evidence of activity when combined with nivolumab[1][5][7].
