Overview
The drug is a **combination therapy of TTI-621 and talimogene laherparepvec**. - **TTI-621 (SIRPα-IgG1 Fc)** is a recombinant fusion protein that acts as a decoy receptor for CD47, blocking the "don't eat me" signal and activating tumor cell phagocytosis by macrophages via its IgG1 Fc region. - **Talimogene laherparepvec (T-Vec, Imlygic)** is an oncolytic immunotherapy derived from a genetically modified herpes simplex virus type-1 (HSV-1) that replicates within tumors and produces the immune stimulatory protein granulocyte-macrophage colony stimulating factor (GM-CSF), inducing direct tumor lysis and promoting local and systemic antitumor immune responses. - The combination seeks to enhance antitumor immunity through both the innate (macrophage activation via CD47 blockade) and adaptive immune responses (T-cell mediated, promoted by oncolysis and GM-CSF expression). This combination has been assessed in patients with unresectable melanoma[3][5].
