TTP054 is a novel, orally available small-molecule agonist of the glucagon-like peptide 1 receptor (GLP-1R), developed for the treatment of type 2 diabetes mellitus. Unlike currently available injectable GLP-1 analogues, TTP054 was designed to be administered orally and acts by binding to a newly identified and patented site on the GLP-1 receptor. In clinical studies, TTP054 demonstrated glucose-lowering effects with reductions in HbA1c and plasma lipids in patients with type 2 diabetes who were not well controlled on other oral anti-diabetic agents. The drug showed good tolerability without significant gastrointestinal side effects or serious adverse events. TTP054 was developed by TransTech Pharma (now vTv Therapeutics) as part of their program to create first-in-class oral non-peptide GLP-1R agonists[1][2][4][5][7].
