Overview

TTP889 is an orally active small molecule that acts as a partial inhibitor of coagulation factor IXa (FIXa), with little or no activity against factors VIIIa, Xa, XIa, or XIIa. It selectively targets the intrinsic coagulation pathway by inhibiting FIXa’s ability to form a fully functional tenase complex with FVIIIa, thereby interfering with fibrin formation and thrombus development. Developed by Transtech Pharma (now vTv Therapeutics), TTP889 was investigated primarily for the prevention of venous thromboembolism (VTE) following hip fracture surgery. Clinical trials demonstrated that while it could inhibit up to 90% of FIXa activity at therapeutic doses and had a favorable bleeding profile, it did not show significant efficacy in extended VTE prophylaxis compared to placebo[1][2][6][8].
