Drug intelligence / Profile preview
TTRA-4201 is a preclinical-stage, ferrous iron-reactive (FIRE) small molecule-drug conjugate developed by Tatara Therapeutics. It consists of the topoisomerase I inhibitor exatecan conjugated to a novel ferrous iron (Fe2+)-reactive trigger group. TTRA-4201 exploits the 'ferro-addiction' of cancer cells, which maintain elevated levels of labile ferrous iron to support rapid growth. Upon entering the tumor microenvironment or cancer cells, TTRA-4201 reacts with intracellular Fe2+, selectively uncaging and releasing active exatecan. This tumor-selective activation mechanism minimizes systemic exposure of free exatecan in peripheral plasma, potentially improving the therapeutic index and reducing toxicities such as neutropenia. TTRA-4201 has demonstrated in vivo stability and robust anti-tumor activity in preclinical colorectal carcinoma xenograft models.
