Overview

TTRNA-DC vaccine is an autologous dendritic cell-based immunotherapy developed by the University of Florida for the treatment of high-grade gliomas, specifically in pediatric populations. The vaccine is produced by extracting total tumor RNA (TTRNA) from a patient's own tumor and pulsing it into the patient's harvested dendritic cells (DCs). These 'primed' DCs are then administered back to the patient, often in combination with granulocyte-macrophage colony-stimulating factor (GM-CSF) as an adjuvant, to stimulate a robust T-cell mediated immune response against a broad spectrum of tumor-specific antigens. This approach aims to overcome the immunosuppressive environment of gliomas and target heterogeneous tumor cell populations. It is currently being evaluated in the ACTION trial in combination with dose-intensified temozolomide and adoptive T-cell therapy (TTRNA-xALT).
