TTX-030 + budigalimab is a combination therapy under clinical investigation primarily for first-line treatment of metastatic pancreatic ductal adenocarcinoma. TTX-030 is a fully human IgG4 monoclonal antibody that acts as a non-competitive allosteric inhibitor of CD39, an ectoenzyme that catalyzes the conversion of extracellular ATP to AMP, a process that facilitates the generation of immunosuppressive adenosine in the tumor microenvironment. By inhibiting CD39, TTX-030 prevents immune suppression by maintaining pro-inflammatory ATP levels and reducing adenosine formation, thereby promoting both innate and adaptive anti-tumor immunity[2][5][6]. Budigalimab is an investigational anti-PD-1 monoclonal antibody, functioning as an immune checkpoint inhibitor to further enhance T cell-mediated antitumor responses. The combination is being evaluated in clinical trials in combination with gemcitabine and nab-paclitaxel chemotherapy[1][3][5].
