Overview

TTX-080 is a first-in-class IgG1 monoclonal antibody that targets HLA-G, an immune checkpoint molecule expressed on various solid tumors. By binding to HLA-G, TTX-080 blocks its interaction with inhibitory receptors ILT2 and ILT4 on lymphoid and myeloid cells, thereby reversing immune suppression in the tumor microenvironment. This blockade reinvigorates CD8+ T cells, enhances natural killer (NK) cell cytolytic activity, and increases macrophage phagocytosis—ultimately promoting anti-tumor immune responses. Developed by Tizona Therapeutics, TTX-080 is being investigated primarily for advanced refractory or resistant solid tumors including metastatic colorectal cancer (mCRC) and head and neck squamous cell carcinoma (HNSCC). Clinical trials have evaluated its safety and preliminary efficacy as monotherapy or in combination with cetuximab or pembrolizumab[1][2][3][4][5][6][7].
