TTX-080 + pembrolizumab

TTX-080 + pembrolizumab is an investigational combination therapy composed of TTX-080, a fully human IgG1 monoclonal antibody and first-in-class antagonist of HLA-G, together with pembrolizumab, an anti-PD-1 checkpoint inhibitor. TTX-080 blocks the immunosuppressive activity of HLA-G (a non-classical MHC class I molecule implicated in tumor immune evasion) by preventing its interaction with inhibitory receptors ILT2 and ILT4 on immune cells, thus restoring anti-tumor immunity. Pembrolizumab blocks the PD-1 receptor, preventing PD-1 mediated T-cell inhibition and sustaining anti-tumor T cell responses. Together, this combination targets two non-redundant immune checkpoints, aiming to enhance anti-tumor immune activity in advanced refractory or resistant solid tumors, including colorectal cancer, head and neck squamous cell carcinoma, non-small cell lung cancer, triple negative breast cancer, renal cell carcinoma, and acral melanoma. Developed by Tizona Therapeutics, this regimen is being evaluated in an open-label, phase 1/1b clinical trial for safety, tolerability, and preliminary efficacy in multiple advanced malignancies[2][3][5][7].
