TTX-114

TTX-114 is a monoclonal antibody developed by Tizona Therapeutics that targets Leukocyte immunoglobulin-like receptor subfamily A member 4 (LILRA4), also known as Immunoglobulin-like transcript 7 (ILT7). ILT7 is a cell surface receptor specifically expressed on plasmacytoid dendritic cells (pDCs), which are the primary producers of Type I interferons (IFN). In autoimmune conditions such as systemic lupus erythematosus (SLE), pDCs are chronically activated, leading to excessive IFN production and subsequent tissue damage. TTX-114 acts as an agonist of the ILT7 receptor; by binding to and cross-linking ILT7, it triggers signaling that suppresses the TLR-mediated production of IFN-alpha and other pro-inflammatory cytokines. This targeted approach aims to modulate the immune response in SLE patients without broad immunosuppression. While Tizona has shifted its primary focus to oncology, TTX-114 remains a notable candidate in its autoimmune portfolio.
