TTX-564

TTX-564 is a first-in-class small molecule inhibitor developed by Trueline Therapeutics that targets the TOX (Thymocyte selection-associated high mobility group box protein) pathway. TOX is a master epigenetic regulator and transcription factor that plays a critical role in the induction and maintenance of T-cell exhaustion, a state of immune dysfunction common in chronic infections and cancer. By inhibiting TOX, TTX-564 aims to reprogram exhausted T-cells, restoring their proliferative capacity and effector functions to enhance the body's anti-tumor immune response. The drug is currently in early clinical development for the treatment of myelodysplastic syndrome (MDS) and various solid tumors, representing a novel approach to immunotherapy by directly addressing the intracellular drivers of T-cell failure.
