TTX-MC138 is an investigational antisense oligonucleotide therapeutic designed to inhibit microRNA-10b (miR-10b), a microRNA implicated as a key driver of metastatic disease in various solid tumors. The drug consists of a synthetic anti-miR10b antagomir conjugated to ultrasmall iron oxide nanoparticles, which can be further chelated and radiolabeled for imaging or tracking purposes. Its mechanism of action involves binding and inhibiting miR-10b, leading to the death of metastatic tumor cells and regression of disease. This approach is hormone receptor independent, potentially allowing treatment across different subtypes of cancers such as breast cancer regardless of ER/PR/HER2 status. Preclinical studies have shown efficacy in models of breast cancer, pancreatic cancer, glioblastoma, and other advanced malignancies. The drug is being developed by TransCode Therapeutics for the treatment of metastatic cancers and is currently in Phase 1/2 clinical trials[2][3][5][6][7][8].
