Overview

TTX-MC138-NODAGA-Cu64 is a radioconjugate drug candidate designed for imaging and potential treatment of metastatic solid tumors. It consists of the microRNA-10b (miR-10b) inhibitor TTX-MC138, which is an anti-miR10b antagomir conjugated to ultrasmall iron oxide nanoparticles, chelated with the chelator NODAGA (1,4,7-triazacyclononane,1-glutaric acid-4,7 acetic acid), and radiolabeled with copper Cu 64. The drug is intended to inhibit miR-10b—a microRNA implicated in cancer metastasis—by delivering the antisense oligonucleotide directly to tumor cells. Its mechanism involves both targeted delivery via nanoparticle technology and molecular inhibition of miR-10b function. The compound has been evaluated in a phase 0/early phase 1 clinical trial for advanced metastatic solid tumors using PET-MRI imaging to assess pharmacokinetics and biodistribution[1][2][4][5][7]. Developed by TransCode Therapeutics, it represents a first-in-class approach targeting metastasis through nucleic-acid based therapeutics.
