TTX-siLIN28B is a small interfering RNA (siRNA) cancer therapeutic candidate developed by TransCode Therapeutics that targets the oncogenic RNA binding protein LIN28B. LIN28B is implicated in tumorigenesis and poor survival outcomes in a variety of cancers, including pancreatic, hepatocellular, colon, lung, multiple myeloma, and neuroblastoma. TTX-siLIN28B utilizes a proprietary nanoparticle delivery platform to deliver siRNA molecules specifically designed to silence LIN28B expression in tumor cells. Preclinical studies have shown that targeting LIN28B with siRNA leads to reduced tumor cell proliferation, induction of cell-cycle arrest, and apoptosis, resulting in potent tumor suppression and inhibition of in vivo tumor xenograft growth. TTX-siLIN28B remains in preclinical development, with an exclusive option agreement for licensing from Massachusetts General Hospital to TransCode Therapeutics[3][1][5][7][8].
