TTX101 is a nanoformed hydrogel-based localized formulation of the natural product piperlongumine being developed by TargTex for the treatment of malignant gliomas, particularly recurrent glioblastoma.[3][5][7][8] The injectable hydrogel is applied intracranially into the resection cavity immediately after tumor excision, where it enables roughly a 200-fold increase in drug load versus bulk material and sustained local release of piperlongumine over several weeks to cover the wound-healing period before radiotherapy.[3][7][10] By delivering high concentrations of piperlongumine directly to the brain, the therapy aims to exploit its tumor-selective cytotoxicity—linked in preclinical models to modulation of oxidative stress and calcium channel–related pathways—to eradicate residual glioma cells while minimizing systemic toxicity.[1][3][7][8] TTX101 has demonstrated reduction in tumor volume and survival benefit in rodent glioblastoma models with minimal local or systemic toxicity, and has received FDA Orphan Drug Designation for malignant gliomas, with a phase 1/2a trial planned as an adjunct to surgery in recurrent glioblastoma.[3][5][7][8]
