TUG1ASO is a novel nucleic acid therapeutic consisting of an antisense oligonucleotide (ASO) targeting the long noncoding RNA (lncRNA) Taurine-upregulated gene 1 (TUG1). It is formulated with a Y-shaped block catiomer drug delivery system, creating a nanoparticle approximately 18 nm in size. TUG1 is highly expressed in glioblastoma and plays a critical role in resolving R-loop formation to maintain tumor cell proliferation. By targeting TUG1, the drug aims to disrupt these processes and inhibit tumor growth. It is currently being evaluated in investigator-initiated Phase I clinical trials for the treatment of recurrent glioblastoma.
