Overview

Tumor-derived vaccine expressing OX40 ligand and 4-1BB ligand is an experimental cancer immunotherapy modality involving tumor cells genetically engineered to express the costimulatory ligands OX40L (TNFSF4) and 4-1BBL (TNFSF9). This approach aims to overcome the immunosuppressive tumor microenvironment by providing direct costimulatory signals to T cells. OX40L interacts with the OX40 receptor (CD134), while 4-1BBL interacts with the 4-1BB receptor (CD137), both of which are members of the tumor necrosis factor receptor superfamily. Preclinical research, including studies by the University of Campinas (UNICAMP), has demonstrated that these vaccines can enhance T-cell proliferation, increase interferon-gamma production, and induce robust antitumor cytotoxicity in models of triple-negative breast cancer and melanoma.
