The tumor fused dendritic cell vaccine (TFDC) is an autologous cellular immunotherapy developed primarily by researchers at The Jikei University School of Medicine for the treatment of aggressive tumors, most notably glioblastoma multiforme (GBM). The vaccine is produced by the electrofusion of patient-derived dendritic cells with autologous tumor cells, creating a hybrid cell that presents a comprehensive repertoire of tumor-associated antigens (TAAs). This approach facilitates the simultaneous presentation of antigens via both MHC class I and class II pathways, thereby activating a robust, polyvalent immune response involving both CD8+ cytotoxic T lymphocytes and CD4+ helper T cells. Clinical investigations have focused on its safety, immunological efficacy, and the identification of prognostic biomarkers, such as HLA-A expression, to optimize patient selection and treatment outcomes.
