Overview

Tumor-infiltrating lymphocyte (TIL) transfer combined with ANV419 is an investigational combination immunotherapy for advanced melanoma and other solid tumors. TIL therapy involves extracting immune cells (lymphocytes) from a patient's tumor tissue, expanding them ex vivo, and reinfusing them to target cancer cells. ANV419 is a novel IL-2/anti-IL-2 antibody fusion protein designed to selectively stimulate CD8+ T cells and natural killer (NK) cells while minimizing activation of regulatory T cells (Tregs). This selectivity is achieved by permitting IL-2 binding to the IL-2 receptor βγ complex but blocking its interaction with the α subunit. The combination aims to enhance anti-tumor immune responses by both increasing effector cell numbers via TILs and further activating these populations through targeted cytokine stimulation with ANV419. Early clinical studies suggest this approach may improve efficacy while reducing toxicity compared to traditional high-dose IL-2 regimens[1][2][3][4][5][6][8].
