Overview

This investigational autologous tumor-infiltrating lymphocyte (TIL) therapy is being developed by Wake Forest University Health Sciences for the treatment of metastatic melanoma. The therapy involves harvesting naturally occurring T cells from a patient's surgically resected tumor tissue, which are then expanded ex vivo using a short-term culture protocol. This approach is a form of adoptive cell therapy designed to harness the patient's own immune system to target cancer. The treatment regimen typically begins with non-myeloablative lymphodepleting chemotherapy (using cyclophosphamide and fludarabine) to create a favorable environment for the infused cells. Following the reinfusion of the expanded TILs, patients receive high-dose aldesleukin (interleukin-2) to support the survival, proliferation, and anti-tumor activity of the lymphocytes in vivo. The TILs are intended to recognize and eliminate melanoma cells through T-cell receptor-mediated recognition of various tumor-associated antigens.
