Tumor lysate-pulsed dendritic cells (TLDCs) are a form of autologous or allogeneic cell-based immunotherapy used in oncology. The process involves harvesting dendritic cells (DCs) from a patient or donor, which are then cultured ex vivo and 'pulsed' or loaded with a whole tumor cell lysate derived from the patient's own tumor or established tumor cell lines. This loading process exposes the DCs to a broad spectrum of tumor-associated antigens (TAAs). Once re-administered to the patient, these mature DCs present the antigens to T cells via MHC class I and II pathways, thereby priming and activating CD4+ and CD8+ T-cell-mediated antitumor immune responses. This platform is primarily investigated in academic settings for various malignancies, including glioblastoma, renal cell carcinoma, and ovarian cancer.
