Overview

Tumor membrane vesicle vaccines (TMV vaccines) are a form of cancer immunotherapy that utilize vesicles derived from the membranes of tumor cells. These vaccines are typically prepared by isolating the plasma membranes from a patient's own (autologous) or allogeneic tumor cells and forming them into nanoscale vesicles. The resulting TMVs display a broad array of native tumor antigens on their surface, which can stimulate an immune response when administered as a vaccine. In some approaches, these TMVs are further engineered to incorporate immunostimulatory molecules such as GPI-anchored B7-1 and IL-12 to enhance their immunogenicity[6][4]. The mechanism of action involves presenting multiple patient-specific or shared tumor antigens to the immune system in their natural conformation, thereby inducing both innate and adaptive anti-tumor immunity. Preclinical studies have shown that TMV vaccines can inhibit tumor growth, improve survival in animal models of head and neck squamous cell carcinoma (HNSCC), induce long-term anti-tumor memory responses, and synergize with immune checkpoint inhibitors like anti-PD1 antibodies in certain models[6][7]. This approach is considered highly personalized when using autologous material.
