A recombinant fusion protein composed of human pro-inflammatory cytokine tumor necrosis factor alpha (TNFα) and the immunostimulatory peptide thymosin alpha 1. This biologic is designed to combine the direct anti-tumor and pro-apoptotic effects of TNFα with the immune-modulating, T cell-maturing, and anti-proliferative properties of thymosin alpha 1. The drug has been investigated primarily for its potential to enhance antitumor immunity in various solid tumors, including breast cancer, melanoma, soft tissue sarcoma, colon cancer, and others. Mechanistically, it acts by stimulating immune effector cells (such as CD4+ and CD8+ T cells), activating natural killer cells, modulating cytokine expression (including ILs and interferons), increasing MHC class I/II expression on target cells for improved immunosurveillance, inhibiting angiogenesis in tumors, and directly suppressing tumor cell proliferation[2][3][8]. Clinical studies have explored its use as an adjunct to chemotherapy or as a neoadjuvant therapy in locally advanced cancers.
