Overview

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) mRNA is an mRNA-based therapeutic designed to induce apoptosis in cancer cells, specifically non-small cell lung cancer (NSCLC). TRAIL is a cytokine that binds to death receptors (DR4 and DR5), triggering the extrinsic apoptotic pathway and subsequent caspase activation. While recombinant TRAIL protein has faced challenges due to rapid clearance and resistance, delivery via mRNA allows for sustained intracellular production of the ligand. In preclinical studies conducted at the University of Alberta, TRAIL mRNA was encapsulated in biodegradable cationic lipopolymers to form nanoparticles. This approach has shown efficacy in reducing viability and inducing mitochondrial depolarization and DNA damage in various NSCLC cell lines, particularly when combined with siRNAs targeting anti-apoptotic mediators to overcome resistance.
