TumorSelect (also known as ART-207 or TumorSelect Paclitaxel) is a preclinical-stage, tumor-targeted paclitaxel prodrug formulation developed by Cloaked Therapeutics in collaboration with Veiled Therapeutics, Arbor Therapeutics, and Monvida Pharmaceuticals. The technology utilizes a lipophilic, acid-labile paclitaxel prodrug incorporated into pseudo-low-density lipoprotein (pseudo-LDL) nanoparticles. This formulation exploits the high cholesterol demand of rapidly proliferating cancer cells, promoting selective uptake of the nanoparticles via low-density lipoprotein (LDL) receptors overexpressed on tumor cells. Once internalized, the acid-labile linker is cleaved in the acidic lysosomal environment of the cancer cell, releasing active paclitaxel to inhibit microtubule depolymerization and induce cell death. This targeted delivery mechanism is designed to enhance therapeutic efficacy while significantly reducing the systemic toxicities (such as myelosuppression and hypersensitivity) associated with conventional paclitaxel formulations like Cremophor EL-based Taxol.
