Overview

**Tumstatin peptide** is a synthetic fragment derived from the N-terminal region (residues 74–98) of tumstatin, itself a cleavage product of collagen IV α3 chain. It acts as a potent and specific **angiogenesis inhibitor** and has demonstrated both antiangiogenic and antitumor properties in preclinical models. Tumstatin peptide exerts its effects primarily by binding to the αvβ3 integrin on proliferating endothelial cells, inhibiting key signaling pathways downstream of integrin engagement, such as FAK, PI3-kinase, Akt, and mTOR, thereby suppressing endothelial cell proliferation and tumor neovascularization[1][4][5][7]. The peptide is shown to work synergistically with anti-VEGF antibody (bevacizumab) for enhanced tumor growth suppression. In diabetic nephropathy models, it inhibits glomerular hypertrophy and related pathological changes[5][7].
