Tunlametinib + nimotuzumab is a combination therapy being evaluated for the treatment of RAS-mutated advanced gastrointestinal malignancies, specifically pancreatic cancer. Tunlametinib (HL-085) is a potent and selective small molecule inhibitor of mitogen-activated protein kinase kinase 1 and 2 (MEK1/2), which are key components of the MAPK/ERK signaling pathway often dysregulated in RAS-mutated cancers. Nimotuzumab is a humanized monoclonal antibody that targets the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling. The combination is designed to achieve dual inhibition of the EGFR and MEK pathways to overcome resistance and enhance anti-tumor efficacy. This specific combination is being investigated in a Phase 2 clinical trial sponsored by the Chinese PLA General Hospital.
