Turletricin (EL219) is a next-generation polyene antifungal and a derivative of amphotericin B, developed by Elion Therapeutics. It is engineered to selectively target fungal ergosterol while avoiding human cholesterol, a design intended to significantly reduce the nephrotoxicity typically associated with traditional amphotericin B. The drug maintains broad-spectrum activity against various fungal pathogens. It has received FDA Qualified Infectious Disease Product (QIDP) and Fast Track designations for the treatment of invasive aspergillosis and early antifungal therapy. Clinical development includes Phase 2 trials for invasive mould infections and cryptococcal meningitis, with a once-weekly dosing regimen.
