Overview

Tusamitamab ravtansine is an antibody-drug conjugate (ADC) designed to target tumor cells expressing carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5). It consists of a humanized monoclonal antibody selective for the extracellular domain of CEACAM5, linked via a cleavable disulfide linker to DM4, a potent cytotoxic maytansinoid payload. Upon intravenous administration, the antibody binds to CEACAM5 on tumor cells and is internalized; intracellular cleavage releases DM4, which inhibits microtubule assembly leading to cell-cycle arrest and apoptosis. The drug was primarily developed for advanced solid tumors with high CEACAM5 expression, especially non-squamous non-small cell lung cancer (NSCLC). Sanofi discontinued its global clinical development in December 2023 after failing to meet primary endpoints in Phase 3 trials for NSCLC[1][2][3][4][6].
